New Compound Shows Promise in Slowing Alzheimer's Progression

Researchers at ETH Zurich have developed 'Compound 10', which may slow Alzheimer's by targeting GRK2 enzyme aggregates.

Researchers at ETH Zurich, led by Ursula Quitterer, have developed an experimental treatment called 'Compound 10' that shows promise in slowing the progression of Alzheimer's disease. The compound has been tested in mice and has demonstrated the ability to significantly slow nerve cell death, a hallmark of dementia, and extend the lifespan of the treated animals.

The research focuses on the enzyme GRK2, which plays a crucial role in cellular regulation. In Alzheimer's patients, an inactive form of GRK2 accumulates in brain cells, forming aggregates that impair mitochondrial function and increase amyloid beta production, a protein associated with Alzheimer's. Compound 10 prevents these aggregates from forming, thereby improving mitochondrial function and reducing amyloid beta levels.

The development of Compound 10 is based on nearly two decades of research, beginning with brain tissue samples from patients in Cairo. The findings were recently published in Cell Reports Medicine. While the compound has shown positive effects in preclinical trials, including benefits to heart function and signs of aging in mice, it has yet to be developed into a treatment for humans.

The researchers have applied for a patent for Compound 10 and are seeking partners to advance it to the next stages of drug development. This new approach targets GRK2, offering a different mechanism from existing Alzheimer's drugs, which could potentially complement current treatments and improve patient quality of life.
